Personally if it was primed I would assemble and then paint. Now if you have several buddies who are willing to stand there and hold the sides while you line up holes and stuff bolts then it may not be so bad on re-assembly. I've replaced bedsides by myself on a long bed and it can be tough and not even attempt it painted. Just my opinion but the only piece I would leave separate is the tailgate
