I believe that the last vent line should go to the fuel evap system in the OE application and through the charcoal canister it would vent the tank. This would be in a system with an unvented gas cap.

The large vent is used to vent the tank during fill up.

You may want to look at some later model Burbans and investigate how they vent their fuel tanks. Maybe you will be able to incorporate some of those components in your truck and enjoy the benefits of a unvented fuel cap.
