Start by getting the book Chevrolet Inline Six-Cylinder Power Manual by Leo Santucci. Most any on-line bookstore has it. There's also a lot of good info at www.inliners.org .
First on my list would be to make it breath better. Stock is under carbed and the exhaust is restricted. Dual carbs and a split exhaust will be a big help. Also an electronic ignition, if you don't have it already. After that you get into cams and head work.

My 250 moves pretty damn good, but it's nowhere near stock. It runs 9.0 to 1 compression, .40 over, Clifford intake, a Holley 390 4 barrel, ported head, and an RV type cam with a header and dual exhaust. It's also been converted to HEI. I can surprise more than a few v8 owners. My truck was built by my uncle before I got it, but feel free to PM me and I'll try to answer any questions I can.
