Quarter panel question ?

I am going to replace one driver side bed side and have a question . I know that there is seam sealer where the bed and quarter panel meet right now . My question is .... do you bolt the panel on and then apply the sealer to keep water from seeping in from the top or does there need to be sealer between the panels before bolting them together . The quarter panel I have does not appear to have any sealer where the two panels meet , only a bead around the top side. I thought that there was some sealer between the panels . Also , what sealer do you recommend for this ? Frank
