New fuel rank and lines

Might be a stupid question, but.
I have installed a new rear mounted fuel tank, new fuel hose and a new fuel filter before the fuel pump in my ’67 stepside. Will the stock 327 engine mounted fuel pump be able to pull fuel from the tank through the new line and filter? The tank has a top mounted fuel outlet and the fuel line is a -6 hose. The truck has not been started for a few months so I know it will have to crank for a while to start, it has always done that even after just a few weeks being idol. Do I need to prime the fuel line or am I just being paranoid?
