Well somewhere down the line a previous owner removed the canister, plugged the vent line, and drill holes in the gas cap Im assuming to serve as the vent. My question in since this worked for the sec 305 I removed will it also work with the 4.8 I am installing in the truck? If so can I just delete the line at the tank and remove the line all together?

Supply and return will obviously stick around. That is pretty creative to drill holes for a vent but a vented cap would be better suited, or just add a proper cap to the vent on the sending unit.

You can run Hardline adapters off the sending unit to convert to AN and then run PTFE hose.
