Check the 8th digit of the vin, if it's a Z it means you have the bi-fuel 5.3 with the notorious fuel pump. Z motors tend to eat fuel pumps like candy, GM is supposedly "working on a fix" but they still havent released a redesigned fuel pump. It's fairly easy to change, all you have to do is remove 8 bolts from the bed, unplug the wiring at the rear, and unbolt the filler neck from the bed, then get 3-4 guys, lift up the bed and slide it backwards, setting the front of the bed on the tires, and the rear of the bed on a 55 gallon drum. should be able to do it under 2 hours that way, if you have a tool box or something heavy in the bed you'll have to drop the tank which doesnt take too long, but is more annoying to do. If your fuel gauge is reading very erratic, and hard to start etc. I would guess you have a Z motor, for the misfire, i'd check the plug/wires on that cylinder, it could be many things. Hard to say without a tech2 hooked up.

Is the code current, or is it a history code? Can you feel the truck missing?

I'd check the plugs and see if they're fouled, also wiggle the wiring harness connectors in the center of the valve cover by the coils, thise wires tend to break pretty often.

It is far more easy to drive the truck till gas tank is low on full, unhook the wires and fuel lines, then undo the 2 straps and drop the tank then to remove the bed, from experience.
