Question Question about the intake manifold gasket

Ok when i bought the gaskets for the intake manifold, it should have 7 square holes, however one of the holes is covered thats the way it came and its at the end, do i need to remove it and make a hole or do i leave it like that, the old gasket did have a hole in there.

forgot to mention that the filled squate does have a little hole in it. guys thanks for the response on other question i had about the antifreeze.
